Gamma Oryzanol - GymBeam | GymBeam.com Skip to main content

Gamma-oryzanol is a substance you can get from rice only if you eat the part most people discard. It is found in rice bran, the layer removed during the production of white rice. One capsule contains 150 mg of gamma-oryzanol—the full daily dose in one go, with no weighing or measuring required. The pack contains 90 capsules, a three-month supply. The product is vegan, gluten-free and lactose-free and tested using the ICP-MS method in an accredited German laboratory—none of the heavy metals tested were detected above the limit of quantification.

Where does gamma-oryzanol come from?

A grain of rice has several layers, and most people consume only the innermost one. On the outside is the husk, which is always removed because it is inedible. Beneath it lies the bran layer, a thin brown coating around the white kernel. This is where most of the rice’s notable components are found. Brown rice retains the bran. White rice loses it. Removing the bran layer is what turns brown rice into white rice, and it is also why white rice can be stored longer and cooks faster. Gamma-oryzanol is found in precisely that removed layer. Rice bran oil is pressed from rice bran and is a common cooking ingredient in Japan; gamma-oryzanol is isolated from this oil.

In other words, it is a substance from the part of rice that industrial processing discards.

What exactly is it?

Despite its name, gamma-oryzanol is not a single substance. It is a group of related compounds, specifically esters of ferulic acid with plant sterols and triterpene alcohols.

Lipids: Two Mechanisms at Once

Gamma-oryzanol is of interest in lipid metabolism because it acts at two completely different sites, a property that follows directly from its structure. The first site is the liver. Molecular studies show that the main components of gamma-oryzanol, cycloartenyl ferulate and 24-methylenecycloartanyl ferulate, bind to the enzyme HMG-CoA reductase. This enzyme initiates cholesterol production in the body and is also the exact target of statins.

Interestingly, these two components do not do the same thing, even though their structures differ only slightly. The first directly reduces the amount of cholesterol in the cell. The second acts primarily on its removal from the cell: it increases the activity of transporters that carry cholesterol out of cells and of the enzyme that converts it into bile acids.¹

The second site is the intestine. The fatty portion of the molecule consists of plant sterols: beta-sitosterol, campesterol and cycloartenol. Their structures are similar enough to cholesterol that they compete with it for absorption. Cholesterol and plant sterols are incorporated into the same micelles in the intestine and use the same transporters. When more sterols are present, less room remains for cholesterol. Plant sterols themselves are absorbed very poorly, so most leave the body, taking cholesterol with them.² Thus, one substance acts on both production and absorption.

Antioxidant action: this is provided by the other half of the molecule

While sterols address fats, ferulic acid provides the antioxidant component. It is a plant phenol, and phenols act in the body as scavengers of free radicals—molecules with an unpaired electron that are therefore unstable and seek to take an electron from something nearby. If they take one from a cell membrane or DNA, damage results. A phenol provides the electron and remains relatively stable itself, stopping the chain reaction.

One aspect of gamma-oryzanol is particularly interesting. Ferulic acid on its own is more soluble in water, but when bound to a sterol, it becomes soluble in fats. This means it can reach places that ordinary water-soluble antioxidants struggle to access: the fatty environment of cell membranes, which is particularly susceptible to oxidative damage. Animal models also describe an effect on the so-called AGE/RAGE axis, a system through which the body responds to proteins damaged by sugar. In a kidney model, gamma-oryzanol reduced markers of inflammation and oxidative stress by suppressing the activity of this receptor.³

Menopause: the oldest trace and also the weakest

Japanese studies from the 1970s and 1980s investigated gamma-oryzanol for menopausal symptoms, and this research led to its registration as a medicine in Japan.⁴ However, its mechanism of action has never been satisfactorily explained. The proposed hypothesis suggests an effect on luteinizing hormone, a signal from the pituitary gland that regulates the activity of the gonads. However, this effect has never been demonstrated in humans and remains speculative.

It is worth noting that older Japanese studies also examined effects on TSH levels, TSH being the hormone that regulates the thyroid gland.⁵ The methods used in these studies are weak by today’s standards, but they show how researchers at the time viewed this substance: as something that affects hormonal regulation in the brain.

Why a capsule?

Gamma-oryzanol is soluble in fat, not in water. This rules out a beverage form and effectively means it is supplied in solid capsules.

Capsules also offer two practical advantages. Each dose is measured during manufacturing: take one capsule and you get exactly 150 mg every time, with no measuring scoop or guesswork. And they are portable: ninety capsules fit in a bag pocket. Because gamma-oryzanol is fat-soluble, it makes sense to take the capsule with a meal containing some fat.

Heavy metals were measured using ICP-MS in accordance with DIN EN 15763—inductively coupled plasma mass spectrometry. This is the most sensitive widely available technique for measuring metals in food; it can detect trace amounts at the parts-per-billion level. The laboratory did not detect any of the metals tested above the limit of quantification.¹⁰

The sample complies with Regulation (EU) 2023/915¹¹ on maximum levels of contaminants in food. This testing is particularly relevant for a product derived from the bran layer of a grain. Bran is the part of the grain most exposed to the external environment, where any contaminants from the soil tend to concentrate more than in the white inner part of the grain.
